چکیده

Construction of the neural netwoks depends largely on precise contacts between neurons and non-neuronal cells. Numerous studies have described different types of adhesive interactions between cells in the nervous system. These include adhesive contacts between neural cell bodies, axonal attachments to glial cells, axon fasciculation, connection between preand postsynaptic specializations as well as to cells outside the nervous system. Although some of the molecules that mediate each of these types of neural adhesive contacts have been characterized, some remain unknown.

LFA-1/ICAM-1 interaction lowers the threshold of B cell activation by facilitating B cell adhesion and synapse formation.

The integrin LFA-1 and its ligand ICAM-1 mediate B cell adhesion, but their role in membrane-bound antigen recognition is still unknown. Here, using planar lipid bilayers and cells expressing ICAM-1 fused to green fluorescence protein, we found that the engagement of B cell receptor (BCR) promotes B cell adhesion by an LFA-1-mediated mechanism. LFA-1 is recruited to form a mature B cell synapse...

The immunological synapse: a molecular machine controlling T cell activation.

The specialized junction between a T lymphocyte and an antigen-presenting cell, the immunological synapse, consists of a central cluster of T cell receptors surrounded by a ring of adhesion molecules. Immunological synapse formation is now shown to be an active and dynamic mechanism that allows T cells to distinguish potential antigenic ligands. Initially, T cell receptor ligands were engaged i...

Structural basis of integrin regulation and signaling.

Integrins are cell adhesion molecules that mediate cell-cell, cell-extracellular matrix, and cell-pathogen interactions. They play critical roles for the immune system in leukocyte trafficking and migration, immunological synapse formation, costimulation, and phagocytosis. Integrin adhesiveness can be dynamically regulated through a process termed inside-out signaling. In addition, ligand bindi...

pH-Sensitive Resist Materials for Combined Photolithographic Patterning of Proteins and Fluid Lipid Bilayers

Photolithography of a pH-sensitive photoresist polymer was performed to pattern both lipid bilayers and proteins onto the same surface. The motivation behind this was to create a substrate mimicking an array of antigenpresenting cells. The substrate would consist of signaling ligand, biotin antiCD3, bound to a lipid bilayer in a regular array of patches. The fluidity of the lipid bilayer would ...

F-actin flow drives affinity maturation and spatial organization of LFA-1 at the immunological synapse

Integrin-dependent interactions between T cells and antigen-presenting cells are vital for proper T cell activation, effector function, and memory. Regulation of integrin function occurs via conformational change, which modulates ligand affinity, and receptor clustering, which modulates valency. Here, we show that conformational intermediates of leukocyte functional antigen 1 (LFA-1) form a con...
